The G25 centrifugal desatling column operates via gel filtration chromatography using a cross-linked dextran matrix. This porous, high molecular weight polymer forms a three-dimensional network that functions as a molecular sieve, enabling separation based on molecular size. Biomolecules larger than the pore size (typically > 5000 Da, such as proteins, peptides, and nucleic acids) are excluded from entering the gel beads. These species migrate through the inter-particle void volume and elute first. In contrast, small molecules including salts, buffer components, and dyes penerate th pores and elute later.
In centrifugal desalting workflows, up to 30% of the column bed volume can be applied as sample. THe G25 centrifugal desalting supports rapid buffer exchange and desalting across multiple while maintaining high recovery of macromolecular analytes.
