Inorganic Pyrophosphatase (E.coli) - 100U

Inorganic Pyrophosphatase (PPase) catalyzes the hydrolysis of inorganic pyrophosphate to form orthophosphate: P2O7-4 +H2O+PPase →2HPO4-2). The enzyme requires a divalent metal cation, with Mg2+ conferring the highest activity. This product exerts its effect at 16 °C - 37 °C, showing the strongest effect at 25 °C. The Inactivation temperature is above 65 °C. 

 

Applications:  

  • High yield RNA synthesis by in vitro transcription. 
  • DNA polymerization reactions: preventing accumulation of pyrophosphate. 
  • Removal of contaminant PPi in reagents used for SNP genotyping by methods based on the detection of pyrophosphate. 

 

Source: PPase is prepared from an E. coli strain containing a clone of the E. coli inorganic pyrophosphatase gene. 

Purity: ≥95% by SDS-PAGE 

Endogenous nucleic acid Concentration: <1 pg/μL by qPCR, 1 U/μL

Definition of Activity Unit: One unit is the amount of enzyme that will generate 1 μmol of phosphate per minute from inorganic pyrophosphate under standard reaction conditions (a 10 minute reaction at 25°C in 100 mM Tris-HCl [pH 8.0], 2 mM MgCl2, 2 mM PPi, reaction volume of 0.5 ml). 

Storage (Dilution) Buffer: 20 mM Tris-HCl, 100 mM NaCl, 1 mM DTT, 0.1 mM EDTA, 50% Glycerol, pH8.0. 

Inactivation or inhibition:
Inhibitors: imidodiphosphate, α ω glycol disphosphates, methanedial diphosphate, 1,2-ethanedial diphosphate.
Inactivation by heating is not complete, reliably removed by spin column or phenol/chloroform extraction.

Store at -20 °C, valid for 12 months.
