Puromycin Solution, 10 mg/mL, Selection Antibiotic for Cell Culture
Puromycin supplied as a ready-made 10 mg/mL solution, so there is no powder to weigh or dissolve. It is added to culture medium to select cells that carry a puromycin resistance gene, for example after transfection or viral transduction.
How it works
Puromycin is an aminonucleoside antibiotic that stops protein synthesis by causing premature termination of the growing peptide chain, which kills cells that are not protected. Cells carrying a puromycin resistance gene (such as pac) survive, so adding puromycin after transfection or transduction leaves a population enriched for the cells that took up your construct.
Key features
- Ready-to-use liquid at 10 mg/mL
- Dilute straight into culture medium, no dissolving step
- Used for selection of puromycin-resistant cells and for establishing stable cell lines
How to use
- Run a kill curve on your untransfected cells to find the lowest concentration that kills them in your culture conditions, since sensitivity varies between cell lines.
- Add puromycin to fresh medium at that concentration after transfection or transduction.
- Replace the medium with fresh puromycin-containing medium as the cells are maintained, and keep a non-resistant control to confirm the selection is working.
