TotaZOL™ Total RNA Purification Reagent

TotaZOL™ Total RNA Purification Reagent is a ready-to-use solution formulated to isolate high-quality total RNA, as well as DNA and proteins, from a wide range of sample types—including human, animal, plant, yeast, and bacterial cells or tissues—within about one hour.
 
This reagent is a monophasic mixture of phenol, guanidine isothiocyanate, and other proprietary components that promote the extraction of both large and small RNA molecules. It preserves RNA integrity by strongly inhibiting RNase activity while simultaneously lysing cells and dissolving cellular components during homogenization.
 
TotaZOL™ Total RNA Purification Reagent enables efficient processing of multiple samples in parallel and represents an enhanced version of the single-step RNA isolation method. From one sample, users can sequentially isolate RNA, DNA, and proteins. After homogenization with the reagent, chloroform is added, which separates the mixture into three phases:
• the clear aqueous phase (containing RNA),
• the interphase (containing DNA),
• the red organic phase (containing proteins).
RNA is precipitated from the aqueous phase with isopropanol. DNA is recovered from the interphase/organic phase with ethanol, and proteins are precipitated from the phenol–ethanol supernatant using isopropanol. Each biomolecule fraction is then washed to remove contaminants and resuspended for downstream use.
 
Applications include:
• RNA: RT-PCR, Northern blotting, dot blot hybridization, poly(A)+ selection, in vitro translation, RNase protection assays, and molecular cloning.
• DNA: PCR, restriction digestion, and Southern blotting.
• Protein: Western blotting, partial recovery of enzymatic activity, and some immunoprecipitation assays.
